A confocal fluorescence microscope with an optico-acoustic beam splitter and a resonance scanner will be acquired to perform live imaging of cultured cells and explanted organs with low fluorescence intensity and under fast imaging conditions. The low fluorescence intensity, the long imaging times (tenths of hours) and the necessity to use low laser power to be compatible with the survival of primary cells sensitive to phototoxicity requires the combination of the best light recovery and the ability to collect the optimal wavelength bandwidth. Moreover, the instrument needs to enable xyzt imaging of deep specimen of at least 50 micrometers deep, with important scattering and low fluorescence. The system needs to be equipped for culture of cells with temperature and CO2 control.
